Volumetric Glue Spore Traps are made by a number of companies,
most of whom offer multiple choices of available cassette types
for a variety of air samplings. Many of the cassettes offered
are better suited for spore trapping airborne Fungals and offer
better methods for analysis as well. The reliance on the Bioaerosol
Cassette Volumetric Glue Spore Trap by the Professionals and

Petri dishes (mould media) using settling methods. Prepared petri

Analytical Methods
Bioaerosol Cassettes: The impaction slide is dyed and direct microscopic
examination performed for presumptive identification, and a count
is preformed on a small portion of the slide. Numbers are then
calculated for reporting count.
Bioaerosol Cassettes can also be read by
Q-PCR DNA; the spore trap can capture a large air volume over
an extended period of time (hours). However, the glue will possibly
dry and lose capture efficiency, which is not that good to start
with.
When results are
compared to viable filter trap taken in tandem with Bioaerosol
Cassettes, comparatively the Bioaerosol Cassettes perform poorly.

Bioaerosol cassette glue spore traps evidently
either do not trap yeast, or it is unrecognizable on a spore
trap. Bioaerosol cassette blow-by was measured utilizing tandem
bioaerosol cassettes and viable filter spore traps or using sterile
2 X 2 gauge in a cassette. The results speak for themselves.
Five different laboratories were used for spore trap analysis.
All spore traps were 150-liter grab samples at 15 liters per
minute 10-minute grab time. No bioaerosol cassette spore traps
reported yeast, yet we recovered yeast from 18 of the two viable
samples.

Yeast is very difficult to recover
from environmental sources and requires liquid culture for most
yeast to grow. The liquid culture must then be converted to solid

Chaetomium and Phoma are underreported because looping
samples on a plate dose not facilitate the growth of Chaetomium
and Phoma.
Chaetomium and Phoma are fungal molds that grow in a Perithecium
(bug like growth), most molds that grow with this morphology
require subsurface environments and may also need Aspergillus
inhibitors as Aspergillus an aggressive filamentous fungi will
early overgrow Chaetomium and Phoma. Double media is necessary
to recover Chaetomium and Phoma when recovering from field samples.
